TADDOL and DIOP Ligands in Asymmetric Titanium and Rhodium Catalysis
TADDOL (α,α,α’,α’-tetraaryl-1,3-dioxolane-4,5-dimethanol) and DIOP (2,3-O-isopropylidene-2,3-dihydroxy-1,4-bis(diphenylphosphino)butane) are two foundational chiral ligands that shaped the modern field of asymmetric catalysis. Although introduced in the 1970s and 1980s, both remain commercially relevant for specific transformations where newer ligands underperform.
TADDOL: The Titanium Workhorse
TADDOL, developed by Seebach, forms chiral titanium alkoxide complexes (typically TADDOLate-TiCl₂ or TADDOLate-Ti(OiPr)₂) that catalyze:
- Diels–Alder reactions — high enantioselectivity with acryloyl and methacryloyl substrates.
- Aldol reactions — addition of ketene silyl acetals to aldehydes.
- Allylations and cyanations — addition of allylstannanes and TMSCN to aldehydes.
- Hetero-Diels–Alder — construction of dihydropyranones.
The four aryl groups of TADDOL create a deep chiral pocket that envelops the titanium center, and the 1,3-dioxolane backbone locks the diol into a rigid conformation.
DIOP: The Phosphine Pioneer
DIOP, introduced by Kagan in 1971, was the first chiral diphosphine to deliver high enantioselectivity in Rh-catalyzed asymmetric hydrogenation of enamides. While DIPAMP and BINAP later surpassed DIOP in selectivity for many substrates, DIOP-Rh remains useful for:
- Enamide hydrogenation — precursor to L-DOPA-type amino acids.
- Asymmetric hydroformylation — with Rh-DIOP for chiral aldehydes.
- Hydrosilylation — of ketones and imines.
The DIOP-Rh chloride dimer is a convenient precatalyst form for rapid screening.
When to Choose TADDOL or DIOP
- TADDOL-Ti when the substrate is a Lewis-acid-activated carbonyl (Diels–Alder, aldol, cyanation).
- DIOP-Rh when screening historical hydrogenation conditions or when DIPAMP/BINAP availability is constrained.
- Both ligands are cost-effective relative to second-generation spiro and biaryl diphosphines.

Historical and Continuing Value
TADDOL and DIOP are the ligands against which all subsequent chiral ligands have been benchmarked. Their documented performance, low cost, and availability in both enantiomeric series keep them on screening decks for new asymmetric transformations, particularly in academic research and early discovery chemistry.
